LCs are dendritic cells (DCs) found in the outermost layer of theskin. DCs are a critical component of the immune system becausethey are the only cells able to 'see' and 'alert' other respondingimmune cells to initiate a protective response against harmfulforeign invaders. Like sentries of the immune system, DCs arestrategically positioned where they are likely to encounter harmfulpathogens. Identifying the source of these specialized immune cellsmay hold exciting possibilities to novel strategies for vaccinationand treatment of autoimmune diseases and inflammatory skindisorders. In contrast to other DCs which are constantly replaced by acirculating pool of bone marrow-derived precursors, LCs has theinteresting ability to maintain themselves throughout life. Whileit is established that these long-lived sentry cells of the skinarise from precursors that are recruited to the skin prior tobirth, this is the first time that the exact origin of theprecursors of LCs is revealed through advanced fate-mappingtechnique (a method of tracing cell lineages to their embryonicorigin). In this study, published in the June issue of Journal of Experimental Medicine, Dr Florent Ginhoux, and his team demonstrated that adult LCsoriginate from two distinct embryonic lineages in two succeedingwaves. The first wave of precursor cells from the yolk sac 'seed'the skin before the onset of the fetal liver. Interestingly, theteam discovered that at the later stage of development, theyolk-sac precursors are largely replaced by a type of white bloodcells from the fetal liver. Said Dr Ginhoux, Principal Investigator of SIgN, "Whether thisunique dual origin of Langerhans cells influences their ability tomaintain skin integrity or dictate their specialized immunefunctions in response to microbes and vaccines needs to beexamined. But having identified their origin surely opens newpossibilities of using them as novel vaccination strategies or astherapeutic tool for treating inflammatory skin diseases likepsoriasis."
