Mesothelioma researchers in Italy have been disappointed in their efforts to fight malignant mesothelioma with an aspirin derivative. A number of studies have suggested that aspirin may play a role in the fight against certain cancers. But the Italian team testing an aspirin derivative called Co-ASS ([2-acetoxy-(2-propynyl) benzoate] hexacarbonyldicobalt) and its chemically-similar analogue Co-EPM (hexacarbonyl [µ-(2-ethylphenyl) methanol] dicobalt) found no significant advantage for mesothelioma. The team tested the two molecules on the two major types of mesothelioma cells, epithelioid and sarcomatoid. The Co-ASS molecule showed the most promise, triggering cell death in the sarcomatoid cells more effectively than Co-EPM or the standard chemotherapy drug for mesothelioma, cisplatin. The molecule worked by inhibiting proteins that are responsible for ensuring that DNA is replicated correctly when cells divide. Unfortunately, neither the Co-ASS nor the Co-EPM molecule had any significant effect on epithelioid cells, which account for the majority of mesothelioma cases worldwide. What they did find is that the molecules were both able to reduce damaging reactive oxygen/nitrogen species in cells, which are byproducts of chronic inflammation. Inflammation caused by long-term irritation from asbestos fibers is believed to be a trigger for mesothelioma. Summarizing their findings for publication, the authors conclude that these molecules make better non-steroidal anti-inflammatories (NSAID) drugs than anti-tumor drugs. Earlier this year, however, a study published in the British Journal of Cancer found that long-term daily use of aspirin had the potential to reduce the size of colon and lung cancer tumors. Further study may still find a role for aspirin in the prevention of mesothelioma, which claims the lives of an estimated 2,500 Americans and thousands more around the world every year.
